Technical Summary: Lactate Analysis (Bulletin ref. TB098720C)

A rapid high performance analysis for lactate in whole blood, plasma or serum, based on the direct reaction with Lactate Oxidase (LOD).

Name:
Codes:

Lactate (II)
GMRD-090/093/092 - Kits for 100/250/4x250 analyser cycles respectively
GMRD-100/103/102 - Kits containing lysing buffer for 100/250/4x250
analyser cycles respectively

Instruments:

All GM7/LM5/GL5 Series analysers

Principle:

In the presence of molecular oxygen, lactate is oxidised by the enzyme Lactate Oxidase to pyruvate and hydrogen peroxide:

Under the conditions of the assay, oxygen consumption is directly proportional to the L-lactate concentration:

Samples:

Intact whole-blood and lysed whole-blood (collected in Analox tubes or capillaries), plasma, neutralised perchloric acid extracts and other aqueous media. Quality Control Serum and sample record cards for attachment and identification of capillaries are available.

Sample volume:
Analysis time:
Precision:

5 µl or 7µl (variable 3.5 - 25 µl)
20 seconds
Typical within-run C.V. 2 % @ 2.5 mmol/L

Linearity:

10 mmol/L (7 µl sample); 90 mg/dl
20 mmol/L (3.5 µl sample); 180 mg/dl

Accuracy:

i) Whole-blood comparison vs YSI 23L:
y (Analox) = 0.98x + 0.055 mmol/L, r = 0.9991, n = 56
ii) Lysed whole-blood vs classical PCA extract spectrophotometric
y (Analox) = 0.99x - 0.05 mmol/L, r = 0.992

Reagent Stability:

Shelf-life 1 year unopened when stored at 0 - 5°C, 60 days after reconstitution, when stored at 0 - 5°C between use. Can be frozen for extended life.

Notes:

i) Lactate methodology has excellent precision at all levels, stable calibration, and is remarkably free from interferences.
ii) Alternative standards at 3.0 and 5.0 mmol/L, and a quality control material, are available in addition to the kit calibrant of 8.0 mmol/L (72.1 mg/dl).
